Hypotensive Effect and Plasma Renin Activity by Acute Administration of Clonidine

Abstract
In order to assess the acute hypotensive effect of clonidine for emergency patients intravenous injection
was used in 31 hypertensive patients, of whom 9 patients were selected out for evaluationof the changes of plasma renin activity.
The results obtained were as follows;
1. With a single dose of 0.075 mg the mean blood pressure reduction in systole was 29 mmHg and in diastole 20mmHg. The corresponding reduction values for 0.15 mg of clonidine were 56 mmHg in systole and 34 mmHg in diastole.
2. Remarkable reduction of diastolic pressure was observed in patients with malignant hypertension with either a single dose of 0.075mg or U. 15mg.
3. When given intravenously, the onset of effect was seen within 5 minutes, its maximum of effect
within 5 to 35 minutes. Duration of effect ran 10 to 190 minutes (average 54 minutes).
4. Frequent side effects were drowsiness and dryness of mouth, but these were mild in severity.
5. The plasma renin activity, determined 3 to 4 hours after injection of clonidine diminished in all of the cases.
6. An effective single dose for intravenous injection was 0.075mg or 0.15mg according to the severity of cases.
